I usually make NTSC 4:3 standard definition animations. I use 720 X 540 (square pixels) in Photoshop to create the artwork, and my After Effects comp is 720 X 540 (square pixels).

This way the graphics look right on the computer screen, and the final product is 720 X 480 (0.9 pixel aspect ratio). And all is right with the world.

But the times, they are a-changin'...

And someone has asked me to do animations for hi-def widescreen NTSC video. What setting do I use in Photoshop and After Effects so that I can use the square pixel aspect ratio? And what setting do I need for the final video output?

Square pixels, 1280x720 (720p), or 1920x1080 (1080p)

You really do need to know the TYPE of HD video you need to deliver before you start the final render.

For example, some 1080 video -- most of it, actually -- is 1440x1080: a 4x3 aspect ratio in square pixels, but this resolution uses a wide pixel aspect ratio to make it 16x9.

Same thing with 720 video: a lot of it is actually 960x720.

Yep. Ask the editor who's getting your stuff. Frame rate, frame size, codec, pixel aspect ratio, delivery medium, progressive or interlaced.

... don't ask the client. ;-) ... unless it's to get the editor's contact information.
