I'm a beginner and need to create a Alpha channel from a separate matte layer which I have imported into AE. How do I add the separate matte as an alpha channel on the fill layer? Thank you.

You can do this by placing the matte layer directly on top of the Fill layer, then set the Track Matte on the Fill layer to Track Luma (or Inverted depending on your matte source).

Another way is to have your matte layer anywhere in the comp and using Channel > Set Matte filter on the Fill layer.
