How can I Roto paint one frame, and than mask a single area of it and duplicate that to other frames?

I don't think I understood the question completely but I usually do the first and the last frame of the roto mask. After that the middle and keep doing it that way because the keyframe will be smart enough to move your mask path or roto brush.
By doing this you save a lot of time and get AE to do some work for you.

I hope that helps.

Let's say I paint out a shadow in just one frame on a wall. If I want to copy the painted wall to other frames, what's a good way to do that? Thanks!

Corey

Can't say anything precisely without seeing the footage. Can you upload a still image to describe what exactly you want to achieve?

If you are using the clone tool(paint effect), select the layer and press "e" to reveal your effects and you should have a paint effect. Twirl that down and there will be a layer for each clone you made. You can move these layers on the timeline to where you want them to start and extend the end points if you need to lengthen the duration.
