I know how to use a gradient layer to control the properties of individual cards, like position, size, rotation, etc. But is there any way to use a gradient layer to control the individual card's color?

That's a job for Photoshop.

Making a bunch of same-sized AE solids in different colors and putting them in a nested comp.

So by making a bunch of same size solids, the idea would essentially be to manually recreate the card dance effect? I had thought about this, but was afraid that if I wanted a lot of cards, say 20 x 20, would that make for very long render time

It wouldn't be any worse than one big picture cut up into a 20x20 array.
