Do I need to transcode or de-interlace HDV footage first.

I want to edit some HDV footage and some footage taken off a DVD. I think I've learnt enough that I need to convert the footage into a different format first, what format? Also should I be de-interlacing the footage or will after effects take care of that for me? I plan on putting the footage onto youtube and DVD, so one is non-interlaced and the other is interlaced. I wasn't sure which line of thought is correct,
1. Leave it in interlaced format and let AE sort and keep track of everything, and their will be no loss in quality from de-interlacing. It will be perfect for the DVD...
2. AE and everything will maybe work better with a progressive video, when doing lots of effects including time remaping stuff, etc.
In either case, what is the best way to do what I want, convert the footage and de-interlace if required. I recall spending hours in the past trying to find a decent de-interlacer. Does anyone know if it is possible to use the VLC media player de-interlacer( or something similar)? From what I recall nothing I've done comes even close to giving the deinterlacing it provides in real time.
For reference the footage is skydiving footage shot with a sony HC3. I'll also be editing footage from a DVD, so I'm not sure if that will just use the same software just different settings or something different.

I ended up getting Lagarith, which is suppose to be a good editing lossless format. I decided to try it out and just put my video into AE, and made movie, just changed settings to this codex. It worked differently than how I expected since it also de-interlaced the video, which was a nice supprise. It looks like in the export settings AE had the field set to none, so it de-interlaced the footage somehow, anyway not sure how but I ended up with decent de-interlaced footage in a decent format.
I'm a little concerned about how good the de-interlace was but from what I can tell its pretty dam amazing....
Is there anything I should know about my mistake that seems to work well.
