If you have only one page, no one can bookmark or get to any of your content except through the home page and your Flash navigation, which can be a downer for the user, or for anyone who wants to send a link or link to a specific part of your content. But if you're happy about that then one page can be nice and neat and slick.

I've done both. It's a nicer user experience to be able to bookmark, and I've found with the pages I've done, the page changes are nearly seamless. I figured out a simple way to pull off the multiple page functionality.
Program your entire site in one flash file. Create your HTML file and duplicate it for each page, and name them accordingly. Then in frame 1 of your movie, have a URL detection script that changes your flash content based on what "page" your on.
---ex: if http://www.yourdomain.com/contact.html, go to my contact content
Then have each of your links call up the actual url, which will automatically load your flash file with the appropriate page showing.

I am just learning Flash CS4, and have been doing the Lynda tutorials for the last couple of days. One of them listed some of the reasons you might choose to go with single, or multi-page (all seem to be content-based decisions):
If you have lots of small info to get to (like Craigslist), a single page will give you links to everything. If you have categories of services (for example, videography, editing, portfolio) you might want to have multiple pages with tabs categorizing your pages. There are also hybrids such as blogs that put everything on one page.
I would suggest you go to tv.adobe.com and check out the free Flash tutorials to see if there's pertinent info there. Next, plunk down 25 bucks for a month's subscription to Lynda.com. You'll find the answer to all your questions there.
