WHen you unzipped the file,. did you click on the StartHere.html file in the top-level directory? The main page gives you and introduction to Twixtor, which you should absolutely read. The "start" page explains where everything is located.

Then click on the "Introduction" button after reading the "Start" page. Then, after that, click on one of the four buttons for After Effects, Premiere Pro, combustion or Final Cut Pro.

(Basically, In the ProjectsAndMovies subdirectory, with all the movies are also some project files:
for AE they are located in the AETwixtorProjects.sit (for Mac, you'll need Stuffit Expander to expand the package of AE projects) or in AETwixtorProjects.zip (for Win) There are also CombustionWorspaces (.sit and .zip), FCPTwixtor project files (one for using the AE-compatible version of Twixtor in FCP, Intel, one for using the AE-compatible version of Twixtor in FCP on PPC Mac, and one using the FxPlug version of Twixtor), and there are similar Premiere Pro files.)

After you unstuff or unzip the appropriate files, you will have projects that allow you to work with the tutorials as a walkthrough guide.
