Hmmm. Your post is a little vague, so we'll probably need more info to really help.
This info would help:
How fast is the hard drive (5400, 7200, 10K)? Is it SATA? IDE? How full is the drive? Is all your media on this drive, with none on your system drive? What resolution is the media you're trying to play back? Is the audio sample rate the same as your project sample rate? How many layers are you trying to play? Any effects? If there are effects, are they rendered?
I'm guessing the drive is too full, it's not fast enough, or it's a combination of the resolution you're trying to play back and the speed of the drive.
