Call it up on the screen.

Hit Command-Shift 4.

Your cursor will turn into a selection target.

Use this to draw a rectangle around whatever you want to capture from the screen.

When you release the mouse button, you'll hear a "click" and the screen grab will show up on your desktop.

Unless you've "remapped" your keyboard somehow - that's it.
