Getting any quality benefit from using Squeeze is going to involve exporting an intermediate file from Premiere that is loseless like Microsoft Uncompressed or QT Animation. Either will be huge files for a DVD, like hundreds of gigs, but for Blu-ray the file sizes will be...well my calculator doesnt go that high!
If you are looking for a step up from AME, look at Main Concept HD plug-in for Premiere CS4. I used the version for CS3 and it worked great and much faster than AME.
