I would get a PC with fast RAM. Run Win7 OS (64 bit).
I would put one of these GPUs in it:
Quadro FX 5800
Quadro FX 4800
Quadro FX 3800
or GeForce GTX 285

I work with a fully loaded (8 core) Mac, AND a fully loaded (4 core) PC, and in AE, the PC beats the Mac on previews and renders. The cost differential was about $8000 more for the Mac. (Full disclosure: I built the PC myself. So some of that is 'assembly' cost.)

As to the "which OS are you more familiar with..." question; you have to ask yourself, "Am I going to be spending most of my time playing with the OS, or the app?" Because with After Effects, the Mac and PC versions look the same once you launch them.
